What makes a character unique?
I would say that a well-written and developed character is unique, regardless of design. It's a lot harder to make a character with a fleshed out personality and backstory, and they're a lot more fun than just a unique design. You could own a white wolf but make them more interesting than all the other white wolves out there. It's kind of like people - some people might be similar on the surface but so much more complicated underneath.
Personally, I tend to have an easier time drawing characters that I can get a feel for. If they're just a blank slate or some fancy design, I get stuck a LOT and usually fall out of the mood to draw them.

Are you interested in one-off characters (normal and abnormal in anatomy) or original species more?
I prefer one-off characters. It makes them more unique. Not everything with extra legs/heads/tails/etc. need to be a species!

Do you prefer designs (for sale) that have pre-made personalities, or do you prefer to make your own?
I like a little bit of both, but sometimes I'm more inspired to write for a character who's a blank slate. If I really like the design and it does have a pre-made personality that I'm not a huge fan of, I could always change the details anyways.

When buying customs, what kind of themes do you usually lean towards?
Fantasy or natural themes. I do like dragons but it's not so easy to find artists willing to draw/design them. ;v;

What's your favorite part of creating characters? Do you design them visually or with words first?
Words. I started out most of my characters with words, and only a few have actually been converted onto a ref sheet later.
Ankh and Thaddaeus were written for long before they actually had set designs. (Ankh was a rather plain-looking lion in his descriptions.)

Do you have a selfish character? One that values themselves over others?
Myrrdin is an extremely selfish individual who prefers to serve himself and only himself. If he has friends, he only uses them to get to higher places. He plays the blame game a fair bit and never believes he's in the wrong. (So much that he sacrificed almost all the people in a kingdom he was meant to be guardian of due to his self-centered habits.)

How about a selfless one? Does this character and your selfish one ever interact?
Cassiel is selfless and spends most of his time watching over others. He puts others before himself and would rather see his friends happy. Sadly, he doesn't interact with Myrrdin; they live in completely different areas.
