Lock Alpha lets you only colour areas which are already coloured - it won't let you erase anything either. You first have to make an area you want to colour and then lock alpha. :]

"Sample All Layers"... Hmm, my explanation will probably be odd, but I'll try.
See how there are blender and "finger" tools? Normally those won't work on empty layers and you have to use them straight on the layer where you've drawn your picture, but when you click the "Sample all layers", you can use those effects on new, "blank" layer, meaning, you don't have to touch the original image and you can adjust the opacity of those effects, or just easily remove the effects if you're not happy with them. :] If this sounds unclear I could try to make a few screenshots of it.
