Do you mean just to colour a picture in the Oekaki?

First, go to the "Color here" layer. If it already has a colour to it, click the "Lock alpha" box and colour in the lineart without worrying about going out of the lines. :3 If the artist has not already coloured the lineart, though, chances are you cannot use the Lock Alpha feature unless the background and the lineart's inside are on different layers.

I hope that helped! ^^ My apologies if I confused you a bit.

First step is to make lines on transparent layer (all new layers you make will automatically be transparent), and make the colouration on other (also transparent layer). Lines- layer needs to be on top of the colouring layer. When you've made the area with brush(or with some other tool) for colouring on the colouring layer, you just click "lock alpha." Then you should be ready for colouring!
Lock Alpha will only let you colour areas that are already coloured. :]
