Lock Alpha only lets you colour areas that are already coloured, it won't let you add (or remove) anything. It makes it easy to colour without going out of the lines. For example, it's used to make colouring easier for the users in Editable Oekakis (or colouring just for yourself easier).
To use lock alpha, you also have to get hang of using layers and transparency.
So, first you make a new layer (because the layer that there is when you first open oekaki is not transparent), make an area with brush/pencil/any tool you want, then click lock alpha. Now, when you colour the area, it will only let you colour things that were already coloured.
